Pyrene, a high molecular weight polycyclic aromatic hydrocarbon (PAH), is a priority pollutant present in soil contaminated with crude oil, coal-tar and complex PAHs. Bacterial consortium CON-3 developed from crude oil contaminated soil of Patiala, Punjab (India) cometabolized 50 µg ml-1 pyrene in the presence of glucose (0.5 %; w/v) at 30 °C, as determined by reverse-phase high performance liquid chromatography (HPLC). Bacillus sp. PK-12, Bacillus sp. PK-13 and Bacillus sp. PK-14 from CON-3, identified by 16S rRNA gene sequence analysis, were able to cometabolize 64 %, 55 % and 53 % of pyrene in 35 days, respectively. With the increase in glucose concentration to 1.0 % (w/v) in growth medium isolates PK-12, PK-13 and PK-14 showed 19 - 46 % uptake of 50 µg... |

ABSTRACT Queen palm is a tropical species of ecological importance and economic potential in various uses, including food. However, the knowledge about the nutritional value of fruits and the estimation of seed viability are still scarce. The aims of this work were to study the biometrics and chemical composition of fruits and seeds; describe the anatomy of embryos and assess the viability of the tetrazolium test. Mature fruits were collected, processed and submitted to biometry, water content, pulp and seeds chemical composition, germination and emergence in the laboratory evaluations. For the tetrazolium test, whole embryos were immersed in solution at 40 °C for one, two, four and six hours and compared with germination. The fruit pulp is rich in... |

ABSTRACT The goal of the present study is to assess the physiological potential of Byrsonima crassifoliaseeds and analyze the procedures of the germination and tetrazolium tests, as well as the conservation of such seeds by testing different combinations of packaging and environment. The experiment used pyrenes of the clone Açu, collected from mother plants in the germplasm collection of muruci plants of Embrapa Amazonia Oriental, in Belém, Brazil. Different temperatures were tested in the germination test (25, 30, 35 and 20 - 30 ºC). The best methodology and assessment dates were determined for the test, and the seedlings of the species were characterized. Viability was assessed with the tetrazolium test, with combinations of hydration methods (by... |

The objective of this study was to investigate the morphology, anatomy and germination behaviour of Phoenix roebelenii seeds. Biometric data were obtained by measuring 100 seeds extracted from recently harvested fruits and air-dried for one day. Four replications of 50 seeds each were previously treated with Vitavax-Thiran and then put to germinate in Sphagnum sp. in plastic trays at room temperature. Morphological details of the seeds were documented with the help of a scanning electronic microscope and then drawings were made with the help of a clear camera coupled to a stereomicroscope. Permanent lamina containing embryo sections were prepared to study its anatomy. The mean dimensions of the seeds were: length of 10.32mm, width of 5.21mm and thickness... |

Tolerance to Polycyclic Hydrocarbons Aromatic (PAHs) is considered an important characteristic when assessing the bioremediation potential of microorganisms. Given this, the objective of this research was to assay filamentous fungi from the Amazon region, isolated from sediments with differents levels of contamination by PAHs, for tolerance to phenanthrene and pyrene. To achieve this, fungal cultures plugs (5 mm), obtained after 7 days growth, were transferred to petri dishes containing 20% Sabouraud dextrose agar medium, after surface innoculation with phenanthrene and pyrene crystals, separately. Radial mycelial growth was evaluated after 10 days at five different concentration levels for each contaminant and control group, all in triplicate for each... |
